results the highest prevalent genotype was genotype 4a, which was found in 57 patients (92%) followed by 2 isolates (3%) with genotype 4o, 2 strains (3%) with genotype 1g and one isolate (2%) with genotype 4n. conclusions this molecular epidemiology study revealed high prevalence of hcv genotype 4, subtype 4a among egyptian patients residing in sharkia governorate, egypt. patients and methods the hcv molecular epidemiology in sharkia governorate was studied using direct sequencing and further phylogenetic analysis of a partial ns5b region of the hcv genome from 63 patients. hcv genotype and subtype were successfully determined in 62 out of 63 patients. background chronic hepatitis c virus infection and its sequela are major health problems facing the egyptian community. the high prevalence and spread rates of the disease require serious actions to stop or decrease these rates. determination of hcv genotypes and subgenotypes adds significant knowledge about the epidemiology of the disease, and provides an added value in the decision making process of what strategy to follow and what therapy response to expect. the molecular epidemiology and genetic variability of hcv variants circulating in egypt still need further analysis. objectives the study was held to evaluate the genotype and subgenotype of the hepatitis c virus circulating in sharkia as one of the large governorates of egypt, which was not included in any study for genotyping of the virus before.

Development of Hepatitis C Virus Genotyping by Real-Time PCR Based on the NS5B Region

BACKGROUND Hepatitis C virus (HCV) genotyping is the most significant predictor of the response to antiviral therapy. The aim of this study was to develop and evaluate a novel real-time PCR method for HCV genotyping based on the NS5B region. METHODOLOGY/PRINCIPAL FINDINGS Two triplex reaction sets were designed, one to detect genotypes 1a, 1b and 3a; and another to detect genotypes 2a, 2b, an...

Phylogenetic and Genotyping of Hepatitis C Virus in Egypt

Objective: Genotyping is considered an important tool for epidemiological and clinical studies. The biological differences between genotypes make genotyping important for decision-making regarding disease management and therapeutic intervention. Methods: This study has established a simple and accurate in house system for HCV core gene typing. The data were confirmed by sequencing and phylogene...

Sequence-based genotyping of hepatitis B virus in general popula-tion

Background: Hepatitis B Virus (HBV) causes acute and chronic liver disease worldwide. HBV has eight genotypes (A to H) which is the reflection of its genome with their characteristic geographical distribution. Each genotype could have different pathogenic and therapeutic characteristics. There have been few records on HBV genotyping in general population from our region. This study aimed to...

Hepatitis C Virus Genotyping in Chronic Hepatitis C Patients

Chronic hepatitis C virus infection is a massive worldwide healthcare burden with estimated costs in the USA alone of over $5 billon per annum. The virus has a 9.5kb positive sense single-stranded RNA genome with striking heterogeneity between isolates, which has led to it being divided into 6 genotypes and more than 50 subtypes and many quasispecies that has been arisen due to the infidelity o...

Seroprevalence of Hepatitis C Virus among Population in Luxor Governorate, Egypt

Hepatitis C virus (HCV), was first identified in 1989 [1], it is a spherical, enveloped, single stranded RNA virus belongs to Flaviviridae family and having humans and chimpanzees as the only species vulnerable to its infection [2,3]. There are at least 7 genоtypes and more than 67 subtypes of HCV were identified, the different HCV genotypes have different distribution [4]. HCV Genotype 4 is th...
